Distinction between signaling mechanisms in lipid rafts vs. caveolae

摘要: The relative importance of lipid rafts vs. specialized termed caveolae to influence signal transduction is not known. Here we show that in cells lacking caveolae, the dually acylated protein, endothelial nitric oxide synthase (eNOS), localizes cholesterol-rich raft domains plasma membrane. In these cells, expression caveolin-1 (cav-1) stimulates biogenesis, promotes interaction cav-1 with eNOS, and inhibition NO release from cells. Interestingly, where does drive assembly, despite equal levels eNOS localization both proteins plasmalemma, physical dramatically less resulting release. Thus, concentrated rafts, closer proximity necessary for negative regulation function, thereby providing first clear example spatial signaling this organelle distinct domains.
