摘要: Studies on the purinergic system normally deal with adenine-based purines, namely, adenine nucleotides and adenosine. However, a guanine-based may also have important neuromodulatory roles. Guanine-based purines exert trophic effects neural cells, protect brain slices in model of hypoxia stimulate glutamate uptake. In vivo, both guanosine 5V-monophosphate (GMP) (GUO) protected against seizures. this study, we investigated if anticonvulsant effect GMP is mediated by or treatments were able to increase adenosine levels. Intraperitoneal (i.p.) 7.5 mg/kg prevented 50% seizures quinolinic acid (QA) increased cerebrospinal fluid (CSF) levels around twofold threefold, respectively; remained unchanged. Intracerebroventricular treatment 960 nmol 80% 5V-nucleotidase inhibitor a-h-methyleneadenosine 5V-diphosphate (AOPCP), when injected 3 min before, reduced 30% protection as well significantly decreased conversion into measured CSF. This study shows that previously reported an seems be related its ability generate through action ecto-5V-nucleotidase. D 2004 Published Elsevier B.V.
