Association with AflR in endosomes reveals new functions for AflJ in aflatoxin biosynthesis.

摘要: Aflatoxins are the most potent naturally occurring carcinogens of fungal origin. Biosynthesis aflatoxin involves coordinated expression more than 25 genes. The function one gene in cluster, aflJ, is not entirely understood but, because previous studies demonstrated a physical interaction between Zn2Cys6 transcription factor AflR and AflJ, AflJ was proposed to act as transcriptional co-activator. Image analysis revealed that, absence aflJ A. parasiticus, endosomes cluster within cells near septa. fused yellow fluorescent protein complemented mutation parasiticus ΔaflJ localized mainly endosomes. We found that co-localizes with both nuclei. Chromatin immunoprecipitation did detect binding at known DNA recognition sites suggesting either does bind these or binds them transiently. Based on data, we hypothesize assists transport from nucleus, thus controlling availability for activation biosynthesis may also assist directing cytoplasmic membrane export.
