Inhibiting Spinal Neuron-Astrocytic Activation Correlates with Synergistic Analgesia of Dexmedetomidine and Ropivacaine

摘要: Background This study aims to identify that intrathecal (i.t.) injection of dexmedetomidine (Dex) and ropivacaine (Ropi) induces synergistic analgesia on chronic inflammatory pain is accompanied with corresponding “neuron-astrocytic” alterations. Methods Male, adult Sprague-Dawley rats were randomly divided into sham, control i.t. medication groups. The profiles Dex, Ropi, their combination detected by Hargreaves heat test investigated the subcutaneous (s.c.) complete Freund adjuvant (CFA) induced in rat was confirmed using isobolographic analysis. During consecutive daily administration, behavior recorded, immunohistochemical staining applied investigate number Fos-immunoreactive (Fos-ir) neurons hour 2 day 1, 3 7, expression glial fibrillary acidic protein (GFAP) within spinal dorsal horn (SDH) 3, 5 7 after s.c. CFA, respectively, then Western blot examine GFAP β-actin levels 7. Results i.t. Dex or Ropi displayed a short-term dose-dependent manner, administrations showed remarkably down-regulated neuronal astrocytic activations indicated decreases Fos-ir SDH, respectively. Conclusion co-delivery shows pain, which “neuron-astrocytic activation” mechanism may play an important role.
