摘要: Rta, an immediate-early protein of Epstein–Barr virus (EBV), is a transcriptional activator that induces lytic gene expression and triggers reactivation. Being located predominantly in the nucleus, Rta can exert its transactivation function through either direct DNA binding or certain indirect mechanisms mediated by cellular signalling other factors. This study examined whether subcellular localization was critical for induction target genes. First, 410KRKK413 identified as nuclear signal (NLS) Rta. An mutant with NLS converted to 410AAAA413 showed cytoplasmic failed activate promoter BGLF5. Interestingly, ectopic still disrupted EBV latency epithelial cell line. Reporter assays revealed NLS-mutated retained ability two promoters, Zp Rp, at considerable level. Thus, could induce endogenous Zta triggering reactivation EBV.
