摘要: This protocol describes the use of solid-phase cytometry for enumeration airborne bacteria and fungi. In contrast with conventional methods, accurate results can be obtained in real time, especially air samples low numbers microorganisms. Air are collected by impaction on a water-soluble polymer that is subsequently dissolved. Part sample filtered over two membrane filters different pore sizes. One filter used to obtain total count all viable microorganisms, second determine number Microorganisms present labeled viability substrate detected quantified using cytometer. The spots microscopically validated an epifluorescence microscope discriminate between bacteria, fungi fluorescent particles. whole procedure takes 5 h complete quantification or high microbial load.
