RAD18 activates the G2/M checkpoint through DNA damage signaling to maintain genome integrity after ionizing radiation exposure

摘要: The ubiquitin ligase RAD18 is involved in post replication repair pathways via its recruitment to stalled forks, and role the ubiquitylation of proliferating cell nuclear antigen (PCNA). Recently, it has been reported that also recruited DNA double strand break (DSB) sites, where plays novel functions damage response induced by ionizing radiation (IR). This new independent PCNA ubiquitylation, but little known about how after IR exposure. Here, we describe a for IR-induced signaling pathway at G2/M phase cycle. Depleting cells reduced factors ATM, γH2AX, 53BP1 foci exposure, attenuated activation checkpoint. Furthermore, depletion increased micronuclei formation death following both vitro vivo. Our data suggest can function as mediator signals activate checkpoint order maintain genome integrity survival
