摘要: A new cell line, designated HDQ-P1, was successfully established from a primary ductal infiltrating mammary carcinoma by using 3T3 feeder layer lethally irradiated to 60 Gy. The HDQ-P1 cells have been grown in culture for over 115 passages and doubling time of hours. Characterization the line performed. This included morphology light transmission electron microscopy, karyotype, growth rate, telomerase expression, tumor antigen xenograft implantation into nude mice, colony formation soft agar, TP53 sequencing, gene copy number C-MYC, C-ERBB-2, C-H-RAS oncogenes. epithelial nature this confirmed ultrastructural analysis, expression cytokeratins, membrane antigen. possess an extensively rearranged polyploid with average 20 recurrent marker chromosomes. Scatchard analysis demonstrated that both were estrogen- progesterone-receptor negative. had same human reverse transcriptase as other breast cancer lines such MDA-MB-231, SK-BR-3, MCF-7. Direct DNA sequencing showed point mutation which yielded stop codon at amino acid 213 exon 6 gene. five-fold amplification C-MYC observed cells. No C-ERBB-2 genes observed. presents unique characteristics may prove be good experimental model investigating biology.
