Interactions of endogenous lipoproteins with capillary endothelium in spontaneously hyperlipoproteinemic rats

摘要: Abstract In spontaneously hyperlipoproteinemic old Sprague-Dawley rats, endogenous lipoproteins (LP) in the size range of 15 to 40 nm were directly visualized within blood vessels due specimen mordanting with tannic acid. LP morphometric analysis at level endothelium diaphragm capillaries revealed that particles dimensions low-density lipoproteins, high-density (HDL1), and very low density occur endothelial structures involved receptor-mediated endocytosis (coated pitsvesicles, endosomes, lysosomes) transcytosis (plasmalemmal vesicles transendothelial channels). No such could be detected intercellular junctions. Intravenously injected cationized ferritin (CF) pI 8.4 bound uniformly forming an CF-LP complex. Examined 5, 20, 60 min after CF administration, complex was found taken up by only (adsorptive via coated fluid phase through a fraction plasmalemmal vesicles). complexes are progressively accumulated lysosomes. These findings reveal importance net surface charge macromolecular for their intracellular sorting fate.
