摘要: Olfactory receptor cells were isolated from the nasal mucosa ofRana esculenta and patch clamped. Best results obtained with free-floating showing ciliary movement. 1)On-cell mode: Current records for up to 50 min. Under control conditions they showed only occasional action potentials. The odorants cineole, amyl acetate isobutyl methoxypyrazine applied in saline by prolonged superfusion. At 500 nanomolar elicited periodic bursts of current transients arising cellular response was rapidly, fully reversibly blocked μm amiloride added odorant solution. With 10 amiloride, partially abolished. 2)Whole-cell Following breakage patch, lost within 5 15 Prior this, evoked a series slow transient depolarizations (0.1/sec, 45 mV peak peak) which reached threshold thus discharge These depolarizing waves stabilized membrane voltage between −80 −90 mV. We conclude that inhibits chemosensory transduction olfactory cells, probably blocking inward pathways open odorants.
