摘要: Adventitious root cultures of Tarenaya rosea were successfully cryopreserved using the encapsulation-vitrification technique. Histological analysis revealed useful information on successive steps cryopreservation. Coupled with complementary histochemical approaches, these studies provided cellular and tissue descriptions T. during cryopreservation contributed to an understanding stress responses, as well characterization anatomical pattern regeneration. The effects exposure duration PVS3 solution (0–120 min), unloading treatment (direct gradual), recovery medium (liquid solid) roots investigated. highest (91%) after cooling in liquid nitrogen (LN) was reached for 90 min, gradual rehydration solution, solid MS medium. showed high multiplication capacity, which maintained up four subcultures. effect structure investigated by histological studies. Plasmolysis intensified loading solutions, but decreased treatment. proportion intercellular spaces increased progressively throughout protocol, culminating cortex disruption. Histochemical analyses polysaccharides, proteins, both lipidic pectic substances spaces. vascular cylinder remained intact, ensuring formation new from pericycle, showing that proliferative capacity had not diminished.
