摘要: Rumex thyrsiflorus Fingerh. is one of the few dioecious plant species that have sex chromosomes. The chromosome constitution females 2n = 12A + XX and 2n = 12A + XY1Y2 males. It a medicinally important has also been object studies on structure function chromosomes ratio. An efficient regeneration protocol was developed from karyologically stable male roots had derived long-term liquid culture. root segments were grown MS medium supplemented with following growth regulators: 2.4-D, NAA, kinetin, BAP TDZ. highest frequency (81.73 %) adventitious shoot formation (16.27 shoots/explant) obtained MS + 0.5 mg/l Regenerated shoots successfully rooted ½ MS + 2 % sucrose + 0.5 mg/l IBA acclimated to in vivo conditions. Histological analysis revealed indirect (via callus) formation. cells morphogenetic callus surrounded by fibrillar similar extracellular matrix. Molecular based genetic markers confirmed all explants male. stability regenerated plantlets using random amplified polymorphic DNA analysis. This first report concerning micropropagation for R. culture, which offers unique opportunity obtain true-to-type plants same sex.
