摘要: The study used Actinidia deliciosa endosperm-derived callus to investigate aspects of the morphology, histology and chemistry extracellular matrix (ECM) structures in morphogenically stable tissue from long-term culture. SEM showed ECM as a membranous layer or reticulated fibrillar granular structure linking peripheral cells domains. TEM confirmed that is distinct heterogeneous layer, up 4 μm thick consisting amorphous dark-staining material, osmiophilic granules fibres present outside outer cell wall. covered surface forming morphogenic domains was reduced during organ growth. This may be linked acquisition competence thus serve structural marker it callus. also observed on senescent contact with area. Treatment living calluses chloroform washing ether–methanol led partial destruction layer. Digestion pectinase removed almost completely exposed strands remnants. protease did not visibly affect Indirect immunofluorescence low-methylesterified pectic epitopes labelled by JIM5 monoclonal antibody. Immunolabelling, histochemistry, solvent enzyme treatments suggested pectins lipids components These compounds indicate protective, water retention and/or communication functions for this external
