Measurement of the (R)- and (S)-isomers of warfarin in patients undergoing anticoagulant therapy.

摘要: An achiral/chiral high-performance liquid chromatographic system for the analysis of total warfarin together with (R)- and (S)-enantiomers in clinical samples has been developed. The achiral is achieved using a C8 column, which coupled to chiral stationary phase, alpha 1-acid glycoprotein (AGP), thereby allowing isomers without interfering serum peaks. A 0.015 M phosphate buffer mobile phase 15% v/v propan-2-ol (pH 7.0) was used on C8/AGP system. UV at 308 nm quantitation column fluorescence (excitation 300 nm, emission 390 nm) detection employed isomer AGP. Retention time 5.95 min, while that (S)- (R)-warfarin AGP 10.38 12.69 respectively. Peak resolution 1.64. All were subjected solid-phase extraction. Data from two patients single dose study indicate two-compartmental model could represent concentration-time data enterohepatic circulation. In some studied during steady state therapy, concentrations (S)-warfarin greater than indicating clearance former slower these patients.
