Colorectal cancer-derived tumor spheroids retain the characteristics of original tumors

摘要: Primary cultures of cancer cells are useful for developing personalized medicine. In this study, we characterized three lines three-dimensional (3D) tumor spheroids established directly from tissues patients with colorectal cancers (CRCs). Each line mainly included EpCAM-positive and expressing putative stem cell markers such as CD133, CD44, CD24, ALDH1, LGR5. These characteristic remained identically months in vitro. Short tandem repeat genotyping suggested that genetic fingerprints these were similar to those the original which they derived. Mutational analysis showed each had same mutation profile APC, KRAS, MLH1, serine-threonine kinase 11, TP53 its parental tissue. One harboring an activating KRAS was resistant cetuximab while remaining two wild-type different responses cetuximab. Immunohistochemical xenograft tumors derived retained histopathological mutational patterns their tumors. Collectively, results clearly 3D generated CRCs preserved characteristics could be used medicines CRCs.
