Re-activated bull spermatozoa: motility, acrosome status and ability to penetrate cervical mucus and zona-free hamster oocytes

摘要: Abstract Semen of four dairy bulls was inactivated by centrifugation twice through a 7% Ficoll solution. After the second centrifugation, sperm fractions were resuspended in 14G buffer and stored for 0, 3, 6 9 days at room temperature. At end each storage period, semen reactivated plugs to final concentration 400 × 106 ml−1 tris-based diluent supplemented with 5 mM caffeine or theophylline. Sperm motility acrosome status assessed during subsequent incubation. penetration into cervical mucus also assessed. The fertilizing ability tested using zona-free hamster oocyte technique. results showed that bovine solution could be beyond Satisfactory proportions motile spermatozoa intact acrosomes recovered after re-activation. inclusion either theophylline level had beneficial effect on percentage (P
