摘要: Summary Objective Synovitis is evident in a substantial subpopulation of patients with osteoarthritis (OA) and associated development pathophysiology. Recently we have shown that adipose-derived stem cells (ASC) inhibit joint destruction collagenase-induced experimental OA (CIOA). In the current study explored role synovitis alarmins S100A8/A9 immunomodulatory capacity ASCs OA. Method CIOA, characterized by synovitis, surgical DMM (destabilization medial meniscus) were treated locally ASCs. Synovial activation, cartilage damage osteophyte size measured on histological sections. Cytokines synovial washouts serum determined using Luminex or enzyme-linked immunosorbent assay (S100A8/A9), mRNA levels reverse-transcriptase (RT)-qPCR. Results Local administration at various time-points (days 7 14) after induction had no effect pathology. At day already 6 h ASC injection expression pro-inflammatory mediators S100A8/A9, interleukin-1beta (IL-1β) KC was down-regulated synovium. IL-1β protein, although low, ASC-treatment CIOA. protein very high 6 48 h decreased ASC-treatment. The protective action treatment CIOA only found when inflammation present time deposition which reflected levels. Finally, successful resulted significantly lower S100A8/A9. Conclusion Our indicates activation rapidly drives anti-inflammatory effects intra-articularly deposited
