Bioprocess efficiency in Rhizopus oryzae lipase production by Pichia pastoris under the control of PAOX1 is oxygen tension dependent

摘要: Abstract Dissolved oxygen (DO) has been stated as a key parameter for Pichia pastoris (Mut+) cultures growing on methanol. DO set point (DOSP) – between 5 and 50% air saturation with time-varying transfer rate (OTR) was investigated during the induction phase of fed batch producing heterologous Rhizopus oryzae lipase (ROL). around 25% found to be best operating condition in terms ROL total productivity, 4-fold increase respect DOSP 5% (minimum value), also corresponded highest specific rates growth (μ), methanol consumption (qS), uptake (qO2), carbon dioxide production (qCO2) activity (qP). However, overall product yield (Y*P/X) obtained 10%, an 45% compared maximal studied (50%). Specific Y*P/X were lower at extremes range tested. The results validate that tension is essential improve bioprocess efficiency enhances reliability system by keeping optimal conditions. Productivity can optimized, thus reducing supply energy requirements, even different target proteins although presenting values.
