摘要: BACKGROUND Exfoliated cervical cell specimens collected in PreservCyt®, a methanol-based medium used ThinPrep® liquid-based cytology, have been archived epidemiologic studies. However, long-term DNA stability and cytologic of these biospecimens not evaluated. METHODS Cervical were into PreservCyt from participants natural history study human papillomavirus (HPV) infection carcinoma Guanacaste, Costa Rica (1993–2000), stored at ambient temperatures. Thirty classified as low-grade squamous intraepithelial lesions by cytology randomly chosen each collection year (except for 1994) selectively assessed molecular stability. Specimens tested 2001 1) HPV the Hybrid Capture 2 test, 2) β-globin polymerase chain reaction amplification multiple length fragments (268, 610, 1327 bp), 3) nuclear preservation visual inspection newly made slides. All testing was done masked to collection. Associations storage time evaluated using standard contingency tables chi-square tests trend. RESULTS Human DNA, detected unaffected time. Stability (PTrend < 0.0001) declined with increasing Approximately 15% could be amplified any fragment after 5 years (collected 1996). In addition, slides 41% rated marginal (32%) or unsatisfactory (9%) 8 1993). CONCLUSIONS Cervical underwent partial degradation several storage. Methodologic studies optimize cells are needed.
