Oxidative Damage, Bleomycin, and Gamma Radiation Induce Different Types of DNA Strand Breaks in Normal Lymphocytes and Thymocytes: A Comet Assay Study

摘要: Most anticancer treatments such as chemo- and radiotherapy induce DNA damage apoptosis in normal cells. The aim of this study was to assess the induction single double strand breaks (ssb dsb, respectively) human lymphocytes rat thymocytes subjected action H2O2, bleomycin ionizing radiation. Normal peripheral young were following treatments: a) H2O2; b) bleomycin, c) gamma-radiation, all with various doses. studied alkaline neutral comet assay for detection ssb dsb. Apoptosis quantified morphologically agarose gel electrophoresis. After H2O2 treatment, a dose-dependent increase observed. Bleomycin treatment produced moderate at lower concentrations striking dsb higher that coincided presence ladders. Gamma radiation initially induced formation ssb, after three hours an manner. laddering appeared only 3 post-irradiation. biomonitoring inflicted by antineoplastic agents can be easily performed could useful monitor modulate radiotherapeutic regimes cancer patients.
