Long-Term Enhancement of Central Synaptic Transmission by Chronic Brain-Derived Neurotrophic Factor Treatment

摘要: Acute effects of neurotrophins on synaptic plasticity have recently received much attention, but the roles these factors in regulating long-lasting changes function remain unclear. To address this issue we studied long-term (days to weeks) and short-term (minutes hours) brain-derived neurotrophic factor (BDNF) excitatory transmission autaptic cultures hippocampal CA1 neurons. We found that BDNF induced enhancement strength non-NMDA receptor-mediated glutamatergic transmission. This upregulation EPSC amplitude occurred via an increase size unitary currents, with no significant contribution from other aspects neuronal electrical including cell size, voltage-gated sodium potassium current levels, number contacts, frequency spontaneous neurotransmitter release. Chronic treatment also decreased degree depression measured response paired stimuli. Thus, both basal use-dependent specific synapse rather than through generalized potentiation growth differentiation. Finally, showed are functionally mechanistically distinct its acute transmission, suggesting that, vivo, activation Trk receptors can different functional depending time course action.
