摘要: Secretion of surfactant proteins A and D (SP-A SP-D) has been difficult to study in vitro because a culture system for maintaining secretion establish. We evaluated several growth factors, corticosteroids, rat serum, fibroblast feeder layer the ability produce maintain polarized epithelium type II cells that secretes SP-A SP-D into apical medium. Type were plated on filter insert coated with an extracellular matrix cultured at air-liquid interface. Keratinocyte factor (KGF) stimulated cell proliferation more than factor-10 (FGF-10), hepatocyte (HGF), or heparin-binding epidermal-like (HB-EGF). Cells presence KGF serum without fibroblasts had high protein mRNA levels exhibited level secretion. Dexamethasone inhibited but increased expression SP-B. In KGF, dexamethasone, mRNAs maintained levels. was found be independent phospholipid
