摘要: The two-element transposon constructs, utilizing either Ac/Ds or Spm/dSpm, allow random tagging of genes in heterologous model species, but are inadequate for directed specific alleles agronomic importance. We propose the use conjunction with Spm/dSpm to develop a four-element system crop-specific alleles. based construct would include both Ds and dSpm along relevant marker function two steps. In first step dSpm(Ds) stocks (a minimum two) be crossed line containing transposases Spm unlinked integrations selected from segregating population by negative selection representing integration at large number locations genome. Selections made which shows partial complete linkage allele interest. second Ac transposase induce transpositions element linked sites thereby exploiting natural tendency jump sites. Unlinked jumps could monitored appropriate genes. proposed interest chromosome addition lines also help efficient genic male sterility systems hybrid seed production tightly marking fertility restorer gene marker.
