Determination of neutral lipids from subcutaneous fat of cured ham by capillary gas chromatography and liquid chromatography

摘要: The determination of neutral lipids in fat cured ham is reported. Fat samples were extracted with chloroform-methanol (2:1) and free fatty acids separated on an aminopropyl minicolumn, the first fraction chloroform-2-propanol (neutral lipids) second 2% acetic acid diethyl ether (free acids). Neutral fractionated minicolumns, silica stationary phases. Two fractions obtained column: (A) triglyceride cholesteryl esters (B) cholesterol mono- diglycerides. Fraction A was applied to column obtain two new fractions: (C) (D) triglycerides. Fractions B C analysed by capillary gas chromatography (cGC) D cGC HPLC. R.S.D.s below 5% except for monoglycerides (8%). Cholesteryl determined 5 min R.S.D. 5%. main triglycerides identified PPO, POS, POO, POL OOO (P = palmitic acid, O oleic L linoleic acid; S stearic). Monoglyceride diglycerides having 18, 34 36 carbon atoms most abundant. HPLC more difficult than because linearity concentration dependent. procedure allowed lipid classes without derivatization
