Anti-melanogenic and anti-oxidant activities of ethanol extract of Kummerowia striata: Kummerowia striata regulate anti-melanogenic activity through down-regulation of TRP-1, TRP-2 and MITF expression.

摘要: Kummerowia striata (K. striata) is used as a traditional medicine for inflammation-related therapy. To determine whether it has beneficial anti-melanogenic and anti-oxidant activities, we investigated the biological activities of ethanol extract (EKS) using variety in vitro cell culture model systems. The activity was assessed B16F10 melanoma cells terms melanin synthesis tyrosinase inhibitory activity. assays were performed 2,2-diphenyl-1-picrylhydrazyl (DPPH) 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS). EKS showed strong DPPH ABTS assays. mRNA transcription levels protein expression tyrosinase, tyrosinase-related 1, 2, microphthalmia-associated factor decreased dose-dependent manner with treatment. Additionally, did not affect viability at different concentrations this study, indicating that mechanism action EKS-mediated inhibition does involve cytotoxicity. Also, confirmed p-coumaric acid quercetin are important compounds anti-melanogenesis antioxidant properties EKS. Collectively, our findings demonstrate first time possesses activities. Further evaluation development functional supplement or cosmetic may be useful skin whitening reducing wrinkles.
