Purification and Partial Characterization of a Basic Protein from Pig Brain

摘要: Abstract A basic protein has been isolated from pig brain. The purification procedure consisted of repeated extractions with chloroform-methanol (2:1 v/v), 10% sodium chloride, and dilute hydrochloric acid. pH 2.4 to 2.6 fraction obtained the acid extraction was further purified by (NH4)2SO4 fractionation, Sephadex G-100 gel filtration, chromatography on Amberlite IRC-50 a gradient (5 20%) guanidinium chloride. All fractions this have examined starch electrophoresis. homogeneity brain established ultracentrifugation, electrophoresis, polyacrylamide end group analysis. Amino analysis performed complete hydrolysate indicated that ratio acidic amino acids is greater than 1.5. An absence cysteine presence high glycine proline content also observed.
