Long-term cultivation of two diploid epithelial cell lines derived from normal rat liver cells

摘要: To obtain a useful rat liver cell line for in vitro carcinogenesis, two diploid epithelial lines were established from 7-day-old male by the repeated colonial clone method. More than 80% of cells each have maintained normal karyotype over 30 months vitro. The identi. fied as conventional Giemsa and trypsin. techniques. They showed little difference morphology growth rate between early late passages. Without cloning, they tended to be heterogenous morphology, became heteroploid in chromosome increased potential with time. Highly heteroploid which derived one produced ascites solid tumors when inoculated into syngeneic rats intraperitoneally. Histologically, diagnosed poorly differentiated hepatocarcinomas. One these passage contained some activity tyrosine transaminase type aldolase .glycokinase. Therefore, it is suggested that represent parenchymal cells.
