摘要: The gene that encodes acetyl-coenzyme A carboxylase (ACCase; EC 6.4.1.2) in the eukaryotic alga Cyclotella cryptica has been isolated and cloned, representing first time a full-length for this enzyme from photosynthetic organism. contains 447-base pair intron is located near putative translation initiation codon 73-base slightly upstream region biotin binding site of enzyme. polypeptide predicted to be composed 2089 amino acids have molecular mass 230 kDa. deduced acid sequence exhibits strong similarity sequences animal yeast ACCases carboxyltransferase domains. There less carboxyl carrier protein domain, although highly conserved Met-Lys-Met present. terminus ACCase characteristics signal sequence, suggesting imported into chloroplasts via endoplasmic reticulum, as shown case certain nuclear-encoded proteins are transported diatom Phaeodactylum tricornutum.
