摘要: Using high molecular-weight proteomic analysis, we previously showed that Staphylococcal nuclease domain-containing protein 1 (SND1) is highly expressed in recurrent androgen-insensitive prostate cancer tissues. SND1 a component of the RNA-induced splicing complex mediates RNA interference, leading to degradation specific mRNAs. The objective this study was further characterize expression and investigate its biological potential cancer. Radical prostatectomy specimens were obtained from 62 patients. immunohistochemical staining patterns evaluated using an in-house polyclonal antibody. We confirmed mRNA cells situ hybridization technique. To determine importance mRNA, knocked down vitro with small interfering observed significant decrease cell growth. 60 cancers (97%), appearing cytoplasm as small, granular structures; it also present at levels specimens, while hyperplasia normal epithelium, weakly or negatively expressed. intensity increased increasing grade aggressiveness As overexpressed cells, growth these suppressed following knockdown vitro, thus representing promising biomarker therapeutic target.
