摘要: Exposure of cells to ionizing radiation (IR) or tumor necrosis factor-alpha (TNF-alpha) results in the stimulation DNA binding activities transcription factors, AP-1 and NF-kappaB. HVH1/CL100, a dual specificity protein phosphatase, may attenuate response by dephosphorylating key upstream element, mitogen-activated kinase (MAPK). The members IkappaB family proteins regulate NF-kappaB response. We examined effects IR TNF-alpha on HVH1 alpha gene expression. Our data demonstrate that treatment head neck squamous carcinoma (PCI-04A) increased steady-state levels mRNAs; however, induction patterns were different. led relatively prolonged mRNAs lasting at least 7 h, while caused transient these expression returned basal within 6 h post-IR treatment. Treatment with cycloheximide did not prevent orTNF-alpha-inducible genes, indicating responses independent new synthesis. These imply phosphatase regulatory factor play important roles cellular TNF-alpha. In addition, kinetics responsiveness indicates mechanisms TNF-alpha-induced signalling are distinct.
