摘要: Purpose: Circulating cell-free DNA (ccfDNA) is a valuable source of tumor material obtained from simple blood sampling that enables noninvasive analysis the genome. Our goal was to carry out multiparametric ccfDNA and evaluate its prognostic value by investigating overall survival (OS) 97 metastatic colorectal cancer patients (mCRC). Experimental Design: Qualitative parameters (determination main KRAS exon2 BRAF V600E mutations) quantitative (total concentration, mutant proportion ccfDNA, integrity index) were determined simultaneously in single run using unique Q-PCR multimarker approach (100% success rate). Results: The median follow-up time 36 months OS 22 months. Patients showing high levels had significantly shorter (18.07 vs. 28.5 months, P = 0.0087). Moreover, multivariate revealed level an independent factor ( 0.034). All interest: with higher mutation loads for detected mutations 0.0089 0.05, respectively). In addition, fragmentation correlated positively decreased exclusive / -mutant cohort 0.0052) appeared as strong 0.0072), whereas it not significant KRAS/BRAF WT 0.67). Conclusions: data provide first qualitative evidence favor mCRC assessment. Clin Cancer Res; 22(12); 3067–77. ©2016 AACR .
