Alteration of spermatogenesis following spermatogonial stem cells transplantation in testicular torsion-detorsion mice.

摘要: Testicular ischemia is the main consequence of testicular torsion, in both clinical and experimental aspects. Preservation auto-transplantation spermatogonial stem cells (SSCs) could be a new treatment for infertility following torsion. To apply idea this study, animals were randomly divided into four groups control, sham, with torsion followed by transplantation (TT). Isolated SSCs from neonatal mice cultured identified flow cytometry (C-KIT−, INTEGRIN β1 +) RT-PCR (Reverse transcription polymerase chain reaction) specific cell markers (Oct4, Gfrα-1, Plzf, Vasa, Itgα 6 , Itgβ 1 ). transplanted upon 2-h TT group. Cultured reperfusion testicle 2 weeks post-testicular Eight weeks after transplantation, SSCs-transplanted testes epididymis removed sperm analysis, weight histopathological evaluation, pre- post-meiotic gene expression assessment qRT-PCR. Our findings indicated that all evaluated parameters (epididymal profile, Johnsen score, Scp-1, Tekt-1 expressions, profile) significantly decreased (group 3) when compared to control group (p ≤ 0.05). However, abovementioned showed significant increase/improvement torsion-transplantation these lower sham up-regulate genes ischemia, which resulted improvement function structure
