摘要: Salmonella typhimurium was detected to levels as low 119 CFUs using the Threshold Immunoassay System. This immunoassay system utilizes solution-based binding of biotin and fluorescein labeled antibodies salmonella, followed by filtration-capture immunocomplex on a biotin-coated nitrocellulose membrane. Lastly, an anti-fluorescein urease conjugate is bound immunocomplex. Detection made possible via silicon chip-based light-addressable potentiometric sensor. In presence urea, converts substrate ammonia CO2 this results in pH change at surface. The resultant monitored with time signal output reported microV s(-1). An experiment whereby chicken carcass washings were fortified salmonella showed recovery 90%, indicating that technique can be used test for under these conditions. Precautions must instrument sample debris will affect flow through membrane hence output.
