HIF-1α activates hypoxia-induced BCL-9 expression in human colorectal cancer cells

摘要: B-cell CLL/lymphoma 9 protein (BCL-9), a multi-functional co-factor in Wnt signaling, induced carcinogenesis as well promoting tumor progression, metastasis and chemo-resistance colorectal cancer (CRC). However, the mechanisms for increased BCL-9 expression CRC were not understood. Here, we report that hypoxia, hallmark of solid tumors, mRNA human cells. Analysis promoter revealed two functional hypoxia-responsive elements (HRE-B HRE-C) can be specifically bound with transactivated by hypoxia inducible factors (HIF) -1α but HIF-2α. Consistently, ectopic HIF-1α HIF-2α transcriptionally levels Knockdown endogenous siRNA largely abolished induction HIF hypoxia. Furthermore, there was strong association specimens. In summary, results from this study demonstrated cells mainly through HIF-1α, which could an important underlying mechanism CRC.
