摘要: Endothelial cells respond to several cytokines by a rapid increase in expression of the adhesion molecules E-selectin and intercellular molecule-1 (ICAM-1), followed gradual decline. The fate these molecules, which was so far unknown, studied. Specific sandwich ELISA for detection soluble (s)E-selectin sICAM-1 were developed. In supernatant, centrifuged 3 hr at 100,000 g remove microparticles, from human umbilical vein endothelial (HUVEC) activated with tumour necrosis factor (TNF), interleukin-1 (IL-1) or lipopolysaccharide (LPS), ICAM-1 could be detected. Biochemical analysis revealed that sE-selectin migrated as band approximately 94,000 MW. amount released directly correlated cell surface expression. Maximal release observed 6-12 after activation HUVEC decreased below limit 24 activation. After activation, gradually increased expression, reached plateau hr, constant days. Since are highly expressed inflammatory sites, resulting high concentrations may affect interactions leucocytes cells. physiological role, however, remains elucidated.
