摘要: ASCT2 is an ASC (alanine-, serine-, cysteine-preferring) neutral amino acid exchanger that may regulate CNS function by transporting substrates including l-serine, l-cysteine, l-glutamine, l-glutamate and d-serine. Despite the potentially important role of in influencing metabolic signaling functions these acids brain, there has been little description its distribution brain tissue. We employed a commercially available human antibody immunohistochemistry studies adult mouse found wide regional for was limited to dendrites labeled anti-microtubule-associated protein-2 cortex, hippocampus striatum. No immunoreactivity observed areas antibodies against neuronal cell body marker (NeuN), or either astrocyte markers, glial fibrillary acidic protein S100β. In cerebellum both Purkinje bodies were positive immunoreactivity. support dendritic localization low affinity (KT > 1 mM), Na+-dependent d-serine l-glutamine uptake characteristic ASCT2-mediated transport P2 synaptosomal preparations. These results suggest be transporter highlight discrepancy between findings tissue culture situ.
