摘要: Background Post-transplant immunosuppression is associated with endothelial dysfunction that may lead to vasculopathy. We have previously demonstrated cyclosporine causes vascular dysfunction. In this we study examined the effect of tacrolimus (Tac) in an identical model. Methods Lewis rats ( n = 8 per group) were injected Tac (low, medium or high dose) saline (Con) daily for 2 weeks. Segments thoracic aorta (TAo) assessed endothelium-dependent (Edep) and -independent (Eind) vasorelaxation (E max ) sensitivity endothelin (ET)-induced vasoconstriction (C ). ET A B receptor (Rc) expression levels determined TAo. Tumor necrosis factor-alpha (TNF-α) transforming growth factor-beta (TGF-β) myocardial (LV) tissue. Plasma tissue oxidative injury quantified by enzyme-linked immunoassay. Results did not impair Edep relaxation when compared Con p 0.69). Impairment sodium nitroprusside–mediated Eind was noted : 69 ± 2%, 54 2%; 0.0001), whereas no such impairment seen diltiazem-mediated 0.06). decreased 222 19%, 162 11%; 0.0002) (Con 0.8 0.1 fmol/ml, 0.4 fmol/ml; 0.02). alter Rc 0.28), but increased Oxidative similar both LV 0.43) Ao 0.73) Similarly, TNF-α 0.16) different between groups, TGF-β a significant decrease treatment Conclusion Our findings suggests has beneficial effects respect function.
