摘要: Abstract Conflicting evidence concerning hepatic amino acid (AA) metabolism in the isolated perfused rat liver (IPRL) led us to investigate response of IPRL using perfusates with various AA contents. Perfusion ( n = 4) whole blood diluted Krebs buffer (1:3, v/v) acute proteolysis on account deprivation, as shown by large release (≈ 1400 μmole 120 min), especially branched-chain (BCAA) (e.g., Leu, 35.4 ± 10.4 nmole·min −1 ·g first hour, 34.3 5.5 second hour). In a attempt prevent proteolysis, livers were previous medium supplemented known for their antiproteolytic activity, at twice physiological concentrations. Results during hour showed uptake several (mainly alanine, glutamine, and proline), reduced BCAA (leucine, 12.5 6.3 ), an increase glucose urea production. However, because use recirculating system, progressive depletion induced reappearance proteolysis. A two-step loading technique, i.e., addition beginning perfusion balanced mixture 60 min caused further decrease 2 hr 6). Under these conditions, most taken up values comparable those observed vivo .
