In situ localization of beta-glucans in the cell wall of Schizosaccharomyces pombe.

摘要: The chemical composition of the cell wall Sz. pombe is known as β-1,3-glucan, β-1,6-glucan, α-1,3-glucan and α-galactomannan; however, three-dimensional interactions those macromolecules have not yet been clarified. Transmission electron microscopy reveals a three-layered structure: outer layer electron-dense, adjacent less dense, third bordering membrane dense. In intact cells pombe, high-resolution scanning microscope surface completely filled with α-galactomannan particles. To better understand organization to complement our previous studies, we set out locate three different types β-glucan by immuno-electron microscopy. Our results suggest that dense contains mainly β-1,6-branched β-1,3-glucan. Occasionally line gold particles can be seen, labelling fine filaments radiating from layer, suggesting some radial contain β-1,6-glucan preferentially located underneath layer. Linear β-1,3-glucan exclusively in primary septum dividing cells. only labels secondary does co-localize linear while present both septa. early stages formation persists until formed; then just before division label disappears. From these involved perhaps separation two daughter addition, frequently found on Golgi apparatus, small vesicles membrane.
