Bacterial Diversity in Subbituminous Coal and Soil from Coal Mine of South Sumatra, Indonesia

摘要: Coal and soil around coal seam environment provide potential factors that support bacterial life. Study of environmental metagenome to determine relationship among the bacteria make up community in layers is reported. DNA extractions were performed three different methods, namely direct extraction, filtration, filtration with blending. The samples subject PCRamplification get V5-V6 16S rRNA gene fragments, then separated by denaturing gradient gel electrophoresis (DGGE). Results showed indirect methods are advantageous can detect more bands compared method. Among tested, a high number ribotypes (Shannon diversity index), but low evenness observed samples. Cluster analysis DGGE mixed clustered separate from its parent, Phylogenetic trees their sequences boomed Firmicutes Actinobacteria It confirmed physico-chemical properties strongly influence evolutionary distance though they little physical distance. This first report which disturbed represented was not artificially reconstructed as microcosm, natural situation virgin adjacent soil.
