摘要: Abstract The functional recovery following non-severing peripheral nerve injury (PNI) is often incomplete. Erythropoietin (EPO) a pleiotropic hormone and it has been shown to protect nerves mild even moderate severity injuries. However, the effectiveness of EPO in severe PNI largely unknown. In this study, we sought investigate neuroprotective effect new dose regimen sciatic crush (SSCI). Adult male mice (8 animals/group) were randomly assigned sham (normal saline, 0.1 ml/mouse), SSCI 0.1 ml/mouse) with (5000 IU/kg) groups. was performed using calibrated forceps for 30 sec. or normal saline administered intraperitoneally immediately after at post-injury day1 2. assessed by function index (SFI), von Frey Test (VFT), grip strength test. Mice euthanized on day 7 21 injury/peri-injury site processed gene (quantitative real-time PCR) protein (immunohistochemistry) expression analysis. significantly improved SFI, VFT, hind limb paw from 7. demonstrated significant regulatory effects mRNA inflammatory (IL-1β TNF-α), anti-inflammatory (IL-10), angiogenesis (VEGF eNOS), myelination (MBP) genes. IL-1β, F4/80, CD31, NF-κB p65, NF-H, MPZ, DHE (redox-sensitive probe) also modulated treatment. conclusion, augments mitigating inflammatory, anti-inflammatory, oxidative stress, angiogenesis, components SSCI.
