Comparative analysis of the karyotype of new human cell line 4BL at long-term cultivation: Ploidy of the chromosomal set

摘要: Long-term cultivation of human cells, including stem can lead to essential transformations the karyotype and genetic instability. The aim this research was a comparative cytogenetic study new cell line 4BL at 160 205 passages. During standard examination, nullisomy monosomy chromosomes 10 13, 4, 8, 11, 15, 17, 21, X; t(1, 11), t (5, 15), t(12, t(16, 21) were observed; also, six regular marker detected. At passages, modal class 42–43 chromosomes. While passaging increased frequency polyploidy cells (from 2.8 36%), disappearance nearhaploid (22.1% 160th passage) decreased level early division chromatids 5 1.5%) observed. We assume stabilization passages consider that it is necessary conduct an additional molecular for objective identification number class, as well chromosomal anomalies, forecasting direction evolution in vitro.
