摘要: FBW7 is a ubiquitin E3 ligase substrate adaptor that targets many important oncoproteins—such as Notch, c-Myc, cyclin E and c-Jun—for ubiquitin-dependent proteolysis. By doing so, it plays crucial roles in cellular processes, including cell cycle progression, growth, metabolism, differentiation apoptosis. Loss of has been observed types human cancer, its role tumour suppressor was confirmed by genetic ablation mice, which leads to the induction tumorigenesis. How exerts suppression function, whether loss de-differentiation or acquisition stemness—a process frequently seen carcinomas—remains unclear. Emerging evidence shows controls stem self-renewal, differentiation, survival multipotency various cells, those haematopoietic nervous systems, liver intestine. Here, we focus on function potential relevance disease therapeutics.
