Human hyaluronidases: electrophoretic multiple forms in somatic tissues and body fluids: Evidence for conserved hyaluronidase potential N-glycosylation sites in different mammalian species

摘要: Abstract Some properties of the multiple forms human hyaluronidases in somatic tissues and body fluids were investigated. Liver placenta exhibited seven hyaluronidase when analyzed electrophoretically on a polyacrylamide–hyaluronan gel. Ovary, breast, myometrium, endometrium, skin, leukocytes platelets displayed distinct patterns enzymatic micropolydispersity. The most acidic synovial fluid serum, some serum an additional basic form. Following sialidase treatment, number decreased to two placenta, three liver broad form serum. native placental remained fully active after thermal inactivation but desialylated was inactivated slowly quickly suggesting higher overall glycosylation plasma enzyme. Potential N-glycosylation sites searched amino acid sequences six several from different mammalian species using PROSITE motif database. A potential site (site 1) with similar tripeptide observed at same position (HYAL1), lysosomes (HYAL2) newly reported (HYAL4 HYALP1). also present mouse (HYAL1) (HYAL2), rat (HYAL2). This absent HYAL3 all sperm (PH-20) studied (human, macaque, mouse, guinea pig, rabbit fox). second location further polypeptide chain. is isoenzymes study whatever organ localization. pattern 2 NVT for except lysosomal origin where it NVS. Such conserved strongly suggest that they may represent actual sites.
