摘要: The present study reports the identification, and characterization of three new putative piscidin paralogues, ecPis-2, ecPis-3 ecPis-4, from orange-spotted grouper (Epinephelus coioides). cDNA piscidins with 207, 216, 231 nt open reading frame encoded respectively a 68-, 71-, 76-amino acid preprotein consisting predicted signal peptide, mature peptide prodomain. phylogenetic analysis indicated that multiple paralogues in one fish species are highly diversified, suggested should be family belonging to superfamily ancient cationic, linear, amphipathic host defence peptides widespread across invertebrate vertebrate taxa comprising insect cecropins ceratotoxins, amphibian dermaseptins. synthetic peptides, ecPis-2S, ecPis-3S ecPis-4S, had strong activities against bacterial fungal species. EcPis-3S exhibited powerful activity infective stage Cryptocaryon irritans, theronts. full length ecPis-2 ecPis-4 by removal ecPis-2L ecPis-4L respectively, potency bacterial, parasitic ecPis-2S was proved exist spleen HPLC followed ESI-LCMS analysis. Basal transcriptions were detected not only potential sites pathogen entry such as gills, skin intestine, but also tissues head kidney, trunk blood cells, abundant immune however different expressed constitutively levels tissues. In addition, expression upregulated challenged Vibrio Parahaemolyticus, at time point after bacteria injection. These results support being important immune-related genes innate system playing multifunctional complementary roles following their structural functional diversification, pattern difference. Finally, this facilitates evaluation 2L, ecPis-3S, -4L templates therapeutic agents pathogens.
