摘要: Catharanthus roseus (periwinkle) produces a wide range of terpenoid indole alkaloids, including several pharmaceutically important compounds, from the intermediate strictosidine. The complete mRNA sequence for enzyme strictosidine synthase (SSS) was determined. Comparison primary structure encoded protein with amino-terminal purified SSS indicated presence signal peptide 31 amino acids in putative translation product. is by single-copy gene indicating that isoenzymes reported others are formed post-translationally single precursor. sss and tryptophan decarboxylase (tdc), encoding another essential alkaloid biosynthesis, coordinately regulated. In plants steady-state levels highest roots. cell suspension cultures genes rapidly down-regulated auxin. contrast, both strongly induced fungal elicitors such as Pythium aphanidermatum culture filtrate or yeast extract. Induction rapid, transcriptional event occurring independent de novo synthesis. These results show first regulatory step complex process leading to accumulation C. cells transcription biosynthetic genes.
