摘要: To further define the nucleic acid determinants that govern recognition of DNA by topoisomerase II, ability enzyme to cleave a 51-base oligonucleotide contained centrally located 19-base hairpin was characterized. Topoisomerase II cleaved 51-mer in site-specific fashion, within hairpin, one nucleotide from 3'-base stem. Protein denaturants were not required trap cleavage products. Although sequence influenced levels enzyme-mediated scission, it did affect spatial location cleavage. scission double-stranded/single-stranded junction at and tail (either single- or double-stranded) least 8 bases length on 5'-side. Cleavage observed when base-pairing eliminated extended produce completely double-stranded substrate. Finally, displayed size constraint for both stem loop structures hairpin. These results indicate is capable recognizing secondary structure acids identifies first structure-specific recognition/cleavage site type enzyme.
