摘要: Thymoquinone, a naturally derived agent, has been shown to possess antioxidant, antiproliferative and proapoptotic activities. In the present study, we explored thymoquinone effects on proteasomal complex, major system involved in removal of damaged, oxidized misfolded proteins. purified 20S complexes, subunit-dependent composition-dependent inhibition was observed, chymotrypsin-like trypsin-like activities were most susceptible treatment. U87 MG T98G malignant glioma cells treated with thymoquinone, 26S proteasome activity measured. Inhibition complex evident both cell lines, but predominantly cells, accompanied by accumulation ubiquitin conjugates. Accumulation p53 Bax, two substrates activity, observed lines. Our results demonstrate that induces selective time-dependent inhibition, isolated enzymes glioblastoma suggest this mechanism could be implicated induction apoptosis cancer cells.
