摘要: Cementum is a unique mineralized connective tissue that covers the root surfaces of teeth. The cementum critical for appropriate maturation periodontium, both during development as well associated with regeneration periodontal tissues, IU; however, one major impediment to understand molecular mechanisms regulate lack markers. Here we report on identification and characterization such differentially human expressed gene, termed "cementum protein-23" (CP-23) appears be ligament cementum-specific. We screened tumor-derived cDNA libraries by transient expression in COS-7 cells "panning" rabbit polyclonal antibody against cementoblastoma conditioned media-derived protein (CP). One isolated cDNA, CP-23, was E. coli antibodies recombinant CP-23 were produced. Expression periodontium examined Northern blot situ hybridization. transcripts cementoblastoma-derived cells, gingival fibroblasts alveolar bone-derived determined RT-PCR. Our results show have 1374-bp containing an open reading frame encodes polypeptide 247 amino acid residues, predicted mass 25.9 kDa represents CP species. cross-reacted type X collagen. Immunoscreening tissues revealed gene product localized cementoid matrix cementoblasts throughout entire surface root, cell subpopulations located paravascularly blood vessels into ligament. Furthermore, 98% putative 15% cultured vitro product. Cementoblastoma contained 5.0 kb mRNA. In hybridization showed strong mRNA cementoblast, around demonstrate novel, tissue-specific-gene being cementoblasts. These findings offer possibility determine cellular events cementogenesis process development. it might provide new venues design translational studies aimed at achieving predictable tissues.
