摘要: Human adipose-derived stromal cells (ASCs) have been shown to possess therapeutic potential in a variety of settings, including cutaneous wound healing; however, it is unknown whether the regenerative properties this cell type can be applied diabetic ulcers. ASCs collected from elective surgical procedures were used treat full-thickness dermal wounds leptin receptor-deficient (db/db) mice. Cells delivered either as multicellular aggregates or suspensions determine impact formulation and delivery methods on biological activity vivo effect. After treatment with that formulated aggregates, experienced significant increase rate closure compared treated an equal number suspension. Analysis culture supernatant gene arrays indicated three-dimensional produce significantly more extracellular matrix proteins (e.g., tenascin C, collagen VI α3, fibronectin) secreted soluble factors hepatocyte growth factor, metalloproteinase-2, metalloproteinase-14) monolayer culture. From these results, clear culture, formulation, method large vitro biology ASCs.
